It's really great work you've done there. But, it just makes me feel depressed since it shows me clearly all the stuff I need is unavailable to me.

great work.
i hope it would include all psychoactive drugs maybe as a table.

it would be great if we had some website,forum,...(or even maybe a thread here) that everyone could write his/her experiences about meds for SAD/Depression and rate their effectiveness and their side effects .

however there is one but is abandoned after it is sold to revolutionhealth.

Good work! Tandospirone and Aripiprazole look particularly interesting, anyone have any experience with them?

Got renewed intrest for NK1 antagonists again, increase in dopamine release by meth/amphetamine and decrease of neurotoxiticy..

Sounds interesting, maybe not as effective as monotherapy, but could be intresting as augmentation agents.

Neurokinin-1 (NK-1) receptor antagonists abrogate methamphetamine-induced striatal dopaminergic neurotoxicity in the murine brain.
Yu J, Cadet JL, Angulo JA.

Department of Biological Sciences, Hunter College of the City University of New York, 695 Park Avenue, New York, NY 10021, USA.
Methamphetamine (METH) is an addictive substance that also causes extensive neural degeneration in the central nervous system. Because METH augments striatal substance P (SP) levels, we hypothesized that this neuropeptide plays a role in methamphetamine-induced toxicity and neural damage in the striatum. In this study we present evidence demonstrating that signaling through the neurokinin-1 (NK-1) receptor by SP plays an important role in methamphetamine-induced toxicity in the striatum. We tested the effects of the selective NK-1 receptor antagonists WIN-51,708 and L-733,060 on several markers of dopaminergic terminal toxicity in the mouse striatum. Administration of NK-1 receptor antagonist prevented the loss of dopamine transporters assessed by autoradiography and western blotting, the loss of tissue dopamine assessed by high-pressure liquid chromatography, and the loss of tyrosine hydroxylase, as well as the induction of glial fibrillary acidic protein determined by western blotting. Pre-treatment with NK-1 receptor antagonist had no effect on METH-induced hyperthermia. Pre-exposure of mice to either of the NK-1 receptor antagonists alone was without effect on all of these neurochemical markers. These results provide the first evidence that tachykinins, particularly SP, acting through NK-1 receptors, play a crucial role in the pathogenesis of nigrostriatal dopaminergic terminal degeneration induced by METH. This finding could lead to novel therapeutic strategies to offset drug addictions as well as in the treatment of a number of disorders including Parkinson's and Huntington's diseases.
Quote:
Blockade of tachykinin NK1 receptors by CP-96345 enhances dopamine release and the striatal dopamine effects of methamphetamine in rats.
Gygi SP, Gibb JW, Johnson M, Hanson GR.

Department of Pharmacology and Toxicology, University of Utah, Salt Lake City 84112.
The nonpeptide, tachykinin NK1 receptor antagonist, CP-96345, permits the study of the physiological role of extrapyramidal substance P systems. Using microdialysis, we observed that locally applied CP-96345 (200 nM) caused a significant increase in dopamine release in the striatum as well as substantially enhancing striatal dopamine release caused by a low dose of methamphetamine (0.5 mg/kg s.c.). In addition, multiple systemic administrations of CP-96345 almost doubled the dopamine-mediated responses of the striatal neurotensin and dynorphin systems to high doses of methamphetamine (10 mg/kg/dose s.c.). Our findings suggest that the physiological role of substance P released in the striatum is to decrease the activity of the nigrostriatal dopamine pathway.
Hmm
Quote:
The effect of intrastriatal administration of LY306740, a specific NK-1 receptor antagonist, on the behavior and changes in gene expression elicited by the psychomotor stimulant, amphetamine, was studied. Acute administration of amphetamine (2.5 mg/kg, i.p.) caused an increase in behavioral activity and preproenkephalin, preprodynorphin and substance P mRNA expression in the striatum. When amphetamine-treated rats were pretreated with LY306740 (35 and 20 nmoles per side, intrastriatally), there was a significant decrease in amphetamine-induced behavioral activity. Quantitative in situ hybridization histochemistry revealed that both concentrations of LY306740 significantly decreased amphetamine-induced mRNA expression of all three neuropeptides. These data indicate that striatal NK-1 receptors modulate amphetamine-induced behavior and mRNA expression of neuropeptides in the rat striatum.

Awesome list rocknroll!

Interesting to see Afobazole on there, the first time I heard about it there wasn't even a wikipedia entry on it. Has anyone here tried it yet?
